The Uptake and Metabolism of Nicotinic Acid by Rabbit Uteri

Abstract
The incubation of rabbit uterine slices-with nicotinic acid^(-7-14C) or nicotinamide^(-7-14C) in the presence of glucose-containing Krebs-Ringer phosphate buffer, (pH7.4) resulted in the disappearance from the medium of nicotinic acid and in the uptake of the vitamin by the uterine slices. In contrast to nicotinic acid, nicotinamide was hardly taken up by the tissue slices.
The uptake of nicotinic acid by the uterine slices was markedly reduced in the absence of glucose or by the presence of sodium cyanide, sodium azide, and 2,4-dinitrophenol in the incubation medium, but did not require ATP added in vitro.
The uptake was not influenced by the pre-treatment of the animal with ovariectomy or by the in vitro addition. of estradiol-17¥â.
About 50 per cent of the nicotinic acid taken up by the tissues remained as such in the tissues, and the remainder was found to be incorporated into nicotinamide mononucleotide, -nicotinic acid mononucleotide and NAD.
These results were interpreted to indicate that only the uterine slices actively take up nicotinic acid and that a considerable portion of the nicotinic acid .taken up is incorporated into NAD via the biosynthetic pathway, which is operating in the erythrocytes.
